Tetrahydrobiopterin (BH4), an essential cofactor for aromatic amine hydroxylases and nitric oxide synthases (NOS), has a critical role in pain sensitivity and persistence. This molecule is up-regulated in dorsal root ganglion cells after nerve injury and can induce behavioral symptoms of pain hypersensitivity in injured rats; inhibiting its synthesis suppresses pain-like behavior. The purpose of my thesis project is to understand how BH4 is involved in the persistence of pain after nerve injury at the molecular level.
