Jonathan Fletcher's research is directed at elucidating molecules within cancer cells that are potential targets for therapeutic intervention. In recent collaborations he has identified:
- A defective form of the enzyme platelet-derived growth factor recepter alpha (PDGFRA), that is associated with many gastrointestinal stromal tumors (GIST).
- A fusion of the proteins BRD4-NUT.
- An Alpha-TFEB fusion in renal tumors.

Jonathan Fletcher received his MD from Boston University. He completed an internship, residency, and chief residency at University Hospital, Boston and fellowships at Dana-Farber Cancer Institute and Children's Hospital Boston and at Brigham and Women's Hospital.
